چکیده

The toll like receptor 4 (TLR4) is expressed in glial cells and reacts to potential toxic entities. This activation triggers various inflammatory reactions. (+)-Naloxone and (+)naltrexone, dextrorotatory enantiomers of opioid receptor antagonists [respectively, (−)naloxone and (−)-naltrexone], have been demonstrated to dock to TLR4 using “in-silico ” models, and function as an antagonist at this site in vivo [1,2]. Recently, (+)-naloxone have been reported to antagonize self-administration of a μ-opioid agonist remifentanil [3]. Further, (+)-naltrexone blocked self-administration of a dopamine uptake inhibitor cocaine [4]. These findings suggested a “novel” TLR4-mediated mechanism underlying the reinforcing effects of drugs of abuse across pharmacological classes. A more recent study [5] has, however, indicated that the TLR4 hypothesis is less viable.

EFFECTS OF CHOLECYSTOKININ RECEPTOR ANTAGONISTS ON MORPHINE- AND COCAINEINDUCED HYPOTHERMIA

The effects of cholecystokinin (CCK) receptor antagonists on hypothermia induced by cocaine or morphine have been studied in mice. In the present work, subcutaneous (SC) injection of cocaine (50-150 mg/kg) or morphine (125-500 mg/kg) induced hypothermia in mice. Administration of CCKA receptor antagonist MK-329 (0.5-1.5 mg/kg), CCKB receptor antagonist L-365, 260 (0.5-1.5 mgl kg) and CCK r...

Lack of self-administration of cocaine in dopamine D1 receptor knock-out mice.

Evidence suggests a critical role for dopamine in the reinforcing effects of cocaine in rats and primates. However, self-administration has been less often studied in the mouse species, and, to date, "knock-out" of individual dopamine-related genes in mice has not been reported to reduce the reinforcing effects of cocaine. We studied the dopamine D1 receptor and cocaine self-administration in m...

Monocyte Expression of Toll-like Receptor-4 in Patients with Stable Angina Undergoing Percutanoeus Coronary Intervention

Background: Toll like receptors (TLRs) are well recognized players in inflammatory conditions. Among them TLR-4 is involved in chronic inflammatory processes such as formation of atherosclerotic plaques. Objective: The present study was aimed to examine the effects of percutanoeus coronary intervention (PCI) as a revascularization method on monocyte expression of hTLR-4 and on the serum levels ...

Stimulants as specific inducers of dopamine-independent σ agonist self-administration in rats.

A previous study showed that cocaine self-administration induced dopamine-independent reinforcing effects of σ agonists mediated by their selective actions at σ1 receptors (σ1Rs), which are intracellularly mobile chaperone proteins implicated in abuse-related effects of stimulants. The present study assessed whether the induction was specific to self-administration of cocaine. Rats were trained...

Evaluating the Effects of Cytomegalovirus Glycoprotein B on the Maturation and Function of Monocyte-derived dendritic cells

Background & Objectives: Interaction of cytomegalovirus glycoprotein B with toll-like receptors of dendritic cells leads to early signaling and innate immune responses. The aim of this study is to evaluate the effects of cytomegalovirus glycoprotein B on the maturation and function of monocyte-derived dendritic cells in treated groups in comparison with control groups. Materials & Methods: Bloo...
